Product Specific Information

The APA5 monoclonal antibody reacts with mouse CD140a (PDGF receptor alpha chain). PDGFRα is a receptor tyrosine kinase that dimerizes upon ligand binding, forming α/α, β/β, or α/β dimers, which act as kinase substrates. CD140a is expressed in embryonic and mesenchymal-derived adult mouse tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on NIH/3T3 cells at ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimal titration recommended.

Fluorophore Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal loss of brightness and FRET efficiency.

Target Information

PDGFRA is a cell surface receptor tyrosine kinase binding platelet-derived growth factors. These growth factors act as mitogens for mesenchymal-origin cells. Receptor dimers (α/α, β/β, or α/β) determine ligand specificity. Knockout mouse studies show PDGFRA is critical for kidney development, as heterozygous mice exhibit kidney defects.
