Product Specific Information

The MR1 monoclonal antibody reacts with mouse CD154, a 39 kDa transmembrane glycoprotein also known as CD40 ligand (CD40L). CD154 is transiently expressed by activated T cells and binds to CD40 on antigen-presenting cells (APC) such as B cells, monocytes/macrophages, and dendritic cells, playing a crucial role in T cell-APC interactions. CD154-CD40 binding transduces signals for T-dependent B cell activation and cell cycle entry.

For flow cytometric staining, stimulation of enriched T cells or CD4 cells is recommended prior to staining with MR1.

Applications Tested: Tested on stimulated mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal titration should be determined empirically.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. A 610/20 bandpass filter is recommended. Ensure instrument compatibility with this fluorochrome.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD40 Ligand (CD40-L, CD154) is a membrane glycoprotein and differentiation antigen expressed on activated T cells. It stimulates B cell proliferation and immunoglobulin secretion in the presence of cytokines, induces cytokine production and tumoricidal activity in monocytes, and co-stimulates T cell proliferation with production of IFN-γ, TNF-α, and IL-2.
