Target Information
The ORF47 gene encodes a multifunctional serine/threonine kinase involved in several processes, including:
- Egress of virus particles from the nucleus
- Modulation of the actin cytoskeleton
- Regulation of viral and cellular gene expression
It regulates nuclear localization of viral envelopment factor proteins 24 and 27 by phosphorylating the protein kinase ORF66, indicating its role in nuclear egress. ORF47 also disrupts host nuclear lamins (LMNA, LMNB1) and phosphorylates the viral Fc receptor composed of glycoproteins E (gE) and I (gI). Phosphorylation of gE by UL13 alters its subcellular localization from the host early endosome to the plasma membrane. ORF47 participates in transcriptional regulation of cellular and viral mRNAs mainly by phosphorylating the viral transcriptional regulator IE63.
