Product Specific Information

The monoclonal antibody eBio440c recognizes Siglec-H, a protein exclusively found on plasmacytoid dendritic cells (pDC) or type I IFN-producing cells (IPC) in naive mice.
Mouse IPC are typically PDCA+, CD11c+, CD11b−, B220+, and Ly-6C+, and respond rapidly to viral infection.

Siglec-H is a transmembrane protein of the Ig superfamily that binds sialic acid but lacks the cytoplasmic ITIM domain. To enable signal transduction, Siglec-H associates with DAP12.

The eBio440c antibody inhibits pDC function (reduces IFNα secretion in response to CpG). Some mouse strains (e.g., SJL) have higher pDC percentages than C57BL/6.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on SJL spleen cells at ≤0.125 µg/test (100 µL final volume; 10⁵–10⁸ cells/test). Optimal dilution should be determined empirically.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-57)
  for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information
