Product Specific Information

The HIM3-4 monoclonal antibody reacts with human CD33, a 67 kDa member of the sialoadhesion family. CD33 is expressed by myelomonocytic precursors, monocytes, mast cells, and granulocytes. Hematopoietic stem cells and lymphocytes do not express this antigen. CD33 plays a role in sialic-acid dependent cell adhesion.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
PerCP-eFluor 710 emits at 710 nm and is excited with the blue laser (488 nm). It can be used in place of PerCP-Cyanine5.5.
Recommended filters: 710/50 bandpass (preferred) or 695/40 (alternative).
Ensure instrument compatibility.

Light Sensitivity: Protect vial and stained samples from light (photo-induced oxidation sensitive).
Fixation: Samples can be stored up to 3 days in the dark at 4°C using:

Target Information

CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases. CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surfaces. It is a therapeutic target for acute myeloid leukemia and a marker for monocytes and granulocyte precursors. CD33 aids in distinguishing myelogenous leukemia from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
