Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. SK7 and UCHT1 antibodies recognize overlapping epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells. Recommended usage: 5 µL (0.8 µg) per test (100 µL final volume).

Cell number per test: 10⁵–10⁸ cells (empirically determined).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample for antibody cocktails or single-color controls.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are encoded on chromosome 11 and are structurally related members of the immunoglobulin superfamily.

• CD3 antigen is present on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes.
• Not expressed on B or NK cells.
• Defects in CD3 genes are associated with CD3 immunodeficiency.
