Description:
The R4-6A2 antibody reacts with mouse interferon-gamma (IFN-γ), a homodimeric 15–20 kDa cytokine secreted by Th1 cells. This antibody functions as a neutralizing antibody. IFN-γ is produced by activated T, B, and NK cells and exhibits antiviral and antiparasitic activity. It synergizes with TNF-α to inhibit proliferation of normal and transformed cells and modulates immune responses across various cell types expressing high-affinity receptors for IFN-γ. Glycosylation does not affect its biological activity.

Applications Reported: Cytokine neutralization
Applications Tested: Verified by LAL assay for low endotoxin levels and tested for IFN-γ bioactivity neutralization.

At 15 ng/mL, the antibody inhibits 50% of the biological effects of 1 ng/mL mouse IFN-γ (ND50) in an EMCV assay of L929 cell protection.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor produced mainly by T-lymphocytes and NK cells in response to antigens and cytokines.
It is a dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits. On SDS-PAGE, it appears as 25, 20, and 15.5 kDa bands due to differential glycosylation.

The biological activity of IFN-γ is species-specific; human IFN-γ does not cross-react with mouse IFN-γ.
Functions include:

  • Antiviral and antitumor activity
  • Induction of MHC class I and II
  • Macrophage activation
  • Enhanced immunoglobulin secretion by B lymphocytes

Activation occurs via IFN-γ receptor I and II, triggering the JAK-STAT pathway.
Human IFN-γ shares ~40% sequence homology with mouse IFN-γ.
Upregulated by IL-2, FGF basic, and EGF; downregulated by vitamin D3 or DMN.
Mutations in the IFN-γ gene are associated with aplastic anemia.
