Product Specific Information
The HIM3-4 monoclonal antibody reacts with human CD33, a 67 kDa member of the sialoadhesin family. CD33 is expressed by myelomonocytic precursors, monocytes, mast cells, and granulocytes. Hematopoietic stem cells and lymphocytes do not express this antigen. CD33 plays a role in sialic acid-dependent cell adhesion.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells.
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume, 10^5–10^8 cells/test).

Target Information
CD33 is a transmembrane protein of the Siglec family with immunoreceptor tyrosine-based inhibitory motifs (ITIMs). It recruits SHP-1 and SHP-2 phosphatases and can undergo ubiquitin-mediated receptor removal. CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues in N- and O-glycans. It is a therapeutic target for acute myeloid leukemia and a marker for peripheral monocytes, helping distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
