Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Phosphorylation at Ser15 is critical for activation and stabilization of p53, enabling it to regulate genes such as p21waf1 involved in G1 or G2/M arrest.

p53 functions to prevent proliferation of damaged cells and acts as an anticancer agent. Alterations in phosphorylation sites (e.g., Ser392) are associated with tumor formation. p53 is also linked to aging, oxidative stress, and cognitive function. While expressed at low levels in normal cells, it is upregulated in transformed cell lines, contributing to malignancy. Mutations in TP53 lead to loss of tumor suppressor activity and are found in many human cancers, including hereditary conditions such as Li-Fraumeni syndrome.
