Description:
Monoclonal antibody QCH6A7 reacts with mouse CD20, a 32-kDa B cell-specific membrane protein expressed from late pro-B to memory B cells. CD20 is involved in intracellular calcium regulation and B cell proliferation. It is not expressed on hematopoietic stem cells or plasma cells. CD20 expression increases with B cell maturation and is not shed or internalized upon antibody binding.
Monoclonal antibodies against CD20 are used to deplete B cells and are effective immunotherapeutic targets in B cell lymphomas and autoimmune diseases such as rheumatoid arthritis and type I diabetes. The QCH6A7 epitope resides within the 44 amino acid extracellular domain of CD20.
Staining with clone QCH6A7 provides improved staining intensity compared to clone AISB12 in flow cytometry.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes and bone marrow cells at ≤1.0 µg/test. A test is defined as the amount of antibody staining a 100 µL cell sample (10⁵–10⁸ cells/test). Titrate for optimal performance.
Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for cytometers with an ultraviolet (355 nm) laser. Ensure instrument compatibility.
Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use:
- Super Bright Complete Staining Buffer (Product # SB-4401-42), or
- Brilliant Stain Buffer™ (Product # 00-4409-75)
to minimize non-specific interactions.
Light Sensitivity:
Protect vial and stained samples from light.
Fixation:

Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, and its localization changes upon BCR aggregation. Unlike many surface proteins, CD20 remains at the membrane after antibody binding and is not internalized or shed, making it an effective target for antibody-mediated B cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
