Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa.
CD16 includes two genes:

  • CD16A (FcγRIIIA): expressed in macrophages and NK cells
  • CD16B (FcγRIIIB): expressed in neutrophils

Two polymorphisms, NA-1 and NA-2, affect phagocytic capacity. CD16 binds IgG immune complexes, showing preference for IgG1 and IgG3. Upon binding, CD16 triggers signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted; tested on human peripheral blood cells at 5 µL (0.25 µg) per test.
A test is defined as the amount of antibody to stain a cell sample in 100 µL final volume (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for cytometers with a 355 nm ultraviolet laser. Confirm instrument compatibility before use.

Buffer Recommendations:
When using multiple polymer dyes (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

  • Super Bright Complete Staining Buffer (Product # SB-4401-42)
  • Brilliant Stain Buffer™ (Product # 00-4409-75)

Target Information

CD16 (FcγRIII) encodes a receptor recognizing the Fc portion of IgG, mediating immune complex clearance and cellular cytotoxicity.

  • Expression: NK cells (membrane-anchored), neutrophils (PI-anchored via FCGR3B)
  • Clinical relevance: Mutations linked to immunodeficiency 20, herpes zoster susceptibility, systemic lupus erythematosus, and alloimmune neonatal neutropenia.
