Product Specific Information

MA1-141 detects collagen I from bovine, human, and porcine. This antibody does not cross-react with collagen types II–XI and does not detect thermally denatured collagen.
Western blot analysis of recombinant bovine collagen I using MA1-141 shows a predominant band around 270 kDa (alpha-1 dimer) and a fainter band at higher molecular weight (alpha-1/alpha-2 trimer).

MA1-141 has been successfully used in ELISA, immunofluorescence, immunohistochemistry (frozen and paraffin), and Western blot procedures.
The epitope is sensitive to routine formalin fixation and paraffin embedding. Strong staining of connective tissue fibers is observed in acetone-fixed or unfixed frozen sections.


Target Information

Collagen I is a Type I collagen with a triple helix structure composed of two alpha-1 chains and one alpha-2 chain.
It is a fibril-forming collagen found in most connective tissues, abundant in bone, cornea, dermis, and tendon.

Mutations in the COL1A2 gene (encoding the alpha-2 chain) are associated with:

  • Osteogenesis imperfecta types I–IV
  • Ehlers-Danlos syndrome type VIIB and classical type
  • Recessive Ehlers-Danlos syndrome
  • Idiopathic osteoporosis
  • Atypical Marfan syndrome

Symptoms from COL1A2 mutations tend to be less severe than COL1A1 mutations, reflecting the different roles of alpha-2 chains in matrix integrity.
Reciprocal translocations between chromosomes 17 and 22 (Collagen Type I genes and PDGFβ gene loci) are linked to dermatofibrosarcoma protuberans, resulting from unregulated PDGFβ expression.
Two transcripts of Collagen I have been identified due to alternate polyadenylation signals.
