Target Information
Glial cell line-derived neurotrophic factor (GDNF) plays key roles in the control of vertebrate neuron survival and differentiation. A novel neurotrophic factor, neurturin (NTN), has been cloned from human and mouse. Physiological responses to neurturin require the presence of receptor tyrosine kinase RET and a glycosylphosphatidylinositol-linked receptor NTNRα (also known as GDNFR-β, RETL2, or GFR-α2). NTN binds to and forms a complex with GFR-α2 and the RET PTK, activating the RET receptor tyrosine kinase pathway. Both NTN and GDNF activate MAP kinase and phosphatidylinositol 3-kinase pathways, playing critical roles in neuronal development. Neurturin and GDNF define a new family of neurotrophic factors.
