Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Murine IFN-gamma. The anti-Murine IFN-gamma-specific antibody was purified by affinity chromatography and subsequently biotinylated.

Sandwich ELISA:
For detection of Murine IFN-gamma, use 0.25–1.0 µg/mL of this antibody in 100 µL/well. In conjunction with PeproTech Polyclonal Anti-Murine IFN-gamma (500-P119) as the capture antibody, this combination allows detection of at least 0.2–0.4 ng/well of Recombinant Murine IFN-gamma.

Western Blot:
For Western blot analysis, use at 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Murine IFN-gamma is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

IFN gamma (Interferon gamma, Type II interferon)
IFN gamma is a macrophage activation factor produced mainly by T-lymphocytes and natural killer cells in response to antigens, mitogens, and other cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

Functions and Characteristics:

• Exhibits antiviral and tumor antiproliferative activity
• Induces MHC class I and II expression
• Activates macrophages and enhances B lymphocyte immunoglobulin secretion
• Species-specific activity (human IFN gamma does not cross-react with mouse)
• Activates the JAK-STAT pathway via IFN gamma receptor I and II
• Upregulated by IL2, FGF basic, and EGF; downregulated by vitamin D3 or DMN
• Mutations in the IFN gamma gene are associated with aplastic anemia
