Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Murine IFN-gamma. Anti-Murine IFN-gamma-specific antibody was purified by affinity chromatography employing an immobilized Murine IFN-gamma matrix.

Sandwich ELISA:
To detect Murine IFN-gamma by sandwich ELISA (100 µL/well antibody solution), use 0.5–2.0 µg/mL of this antibody. When used with PeproTech Biotinylated Anti-Murine IFN-gamma (500-P119Bt) as a detection antibody, it allows detection of 0.2–0.4 ng/well of Recombinant Murine IFN-gamma.

Western Blot:
For Western Blot analysis, use at 0.1–0.2 µg/mL. Detection limit is 1.5–3.0 ng/lane under reducing or non-reducing conditions, when used with compatible secondary reagents.

IFN gamma (Interferon gamma, Type II interferon)

• Macrophage activation factor and immune interferon produced by T-lymphocytes and NK cells.
• Dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits.
• Exhibits species-specific biological activity; human IFN gamma does not cross-react with mouse.
• Functions: antiviral and antiproliferative activity, MHC class I/II induction, macrophage activation, and enhanced B-cell immunoglobulin secretion.
• Involved in cytokine regulation and activates the JAK-STAT pathway via IFN gamma receptor I and II.
• Approximately 40% sequence homology between human and mouse IFN gamma.
• Upregulated by IL-2, FGF basic, EGF; downregulated by vitamin D3 or DMN.
• IFN gamma gene mutations are associated with aplastic anemia.
