The IK22/5 monoclonal antibody reacts with mouse CD254, also known as TRANCE (TNF-related activation-induced cytokine receptor) and RANKL (receptor activator of NF-kappa B ligand), or OPGL (Osteoprotegerin Ligand).
TRANCE is a type II membrane protein (~35 kDa) closely related to TRAIL, FasL, and TNF. TRANCE mRNA is upregulated by TCR stimulation and expressed abundantly by mouse T cells in thymus and lymph nodes, but not by B cells.
TRANCE-deficient mice lack osteoclasts and show defects in T-dendritic and T-B cell interactions, critical for osteoclast differentiation. The TRANCE-RANK interaction regulates anti-apoptotic molecule Bcl-xL expression.

Target Information

This gene encodes a TNF cytokine family member that acts as a ligand for osteoprotegerin and is essential for osteoclast differentiation and activation.
It functions as a dendritic cell survival factor and regulates T cell-dependent immune responses.
Activation of T cells induces expression of this gene, leading to increased osteoclastogenesis and bone loss.
It activates antiapoptotic kinase AKT/PKB via a signaling complex involving SRC kinase and TRAF6.
Gene disruption in mice results in severe osteopetrosis, lack of osteoclasts, and defects in T/B lymphocyte differentiation and mammary gland development.
Two alternatively spliced transcript variants have been identified.
