Target Information
Sav1 is a regulator of STK3/MST2 and STK4/MST1 in the Hippo signaling pathway, which is crucial for organ size control and tumor suppression by restricting proliferation and promoting apoptosis.
The pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, in complex with SAV1, activate LATS1/2 and MOB1, which phosphorylate and inactivate YAP1 and WWTR1/TAZ.
Phosphorylation of YAP1 prevents its nuclear translocation, regulating genes linked to cell proliferation, death, and migration. SAV1 is essential for activation of STK3/MST2 and STK4/MST1, promoting cell-cycle exit and differentiation in epithelial tissues.
It also regulates centrosome disjunction through NEK2 localization and phosphorylation of CROCC and CEP250. In conjunction with STK3/MST2, Sav1 activates ESR1 transcriptional activity via phosphorylation modulation.
