Product Specific Information

  • Antibody is tested in direct ELISA.
  • For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

GAD-65 and GAD-67 are members of the group II decarboxylase family of proteins and catalyze the rate-limiting step in GABA (gamma-aminobutyric acid) synthesis from L-glutamic acid.
Both GADs are expressed in the brain:

  • GAD-65 localizes to synaptic vesicle membranes in nerve terminals.
  • GAD-67 is distributed throughout the cell and responsible for basal levels of GABA synthesis.
    During increased neurotransmission demand, GAD-65 transiently activates to assist GABA production.
    Loss of GAD-65 impairs GABA neurotransmission, while loss of GAD-67 is lethal.
    Alternative splicing produces two isoforms: the predominant GAD-67 and the minor GAD-25 form.
