Description:
The monoclonal antibody 1A4 recognizes human, mouse, and rat alpha-smooth muscle actin (α-SMA), a 42 kDa cytoskeletal protein expressed in smooth muscle cells, myofibroblasts, and myoepithelial cells. It is useful for identifying leiomyomas, leiomyosarcomas, and pleomorphic adenomas, and serves as a prognostic marker for basal cell carcinoma.

Applications Reported:
The 1A4 antibody has been reported for use in:

• Intracellular staining followed by flow cytometric analysis
• Immunoprecipitation
• Western blotting
• Immunohistochemistry (frozen and paraffin-embedded tissues)
• Microscopy and immunocytochemistry

Applications Tested:

• IHC (FFPE human tissue): ≤1 µg/mL
• ICC (formaldehyde or methanol-fixed cells): 1 µg/mL
• WB (reduced rat and mouse intestine): ≤5 µg/mL
  Antibody concentration should be optimized for each assay.

Smooth Muscle Actin (α-SMA) is a member of the actin protein family involved in cell structure, motility, and contraction.

• Alpha actin: major component of the contractile apparatus
• Beta and gamma actins: involved in cell motility regulation

Smooth muscle actin is found in vessel walls, gut wall, myometrium, and myoepithelial cells of breast and salivary glands.
Mutations in the smooth muscle actin gene are linked to familial thoracic aortic aneurysm type 6.
Alpha-smooth muscle actin is abundant in vascular and visceral smooth muscle cells and appears in fibroblastic stress fibers during wound healing and fibrocontractive diseases.
