Product Specific Information
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with CD3 gamma and delta, the epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.
Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometry of normal human peripheral blood cells (≤1 µg/test).
A test is defined as the amount of antibody staining a sample in 100 µL volume. Cell number may range from 10⁵–10⁸ cells/test. Optimize concentration for best performance.

Alexa Fluor™ 700 emits at 723 nm, excited by red laser (633 nm).
Recommended filter: 685 LP mirror and 710/20 filter.

Target Information
The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are immunoglobulin superfamily members encoded on chromosome 11. These subunits associate with cytoplasmic signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Altered expression may occur in autoimmune diseases. Mutations in CD3 genes are associated with CD3 immunodeficiency.
