Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3ε, a 20 kDa subunit of the TCR complex. Along with the other CD3 subunits (γ and δ), the ε chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling, resulting in cellular activation and proliferation.

  • Applications Tested: Pre-titrated and tested using normal human peripheral blood cells
  • Recommended Usage: 5 µL (0.5 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test)
  • Fluorochrome Details: Alexa Fluor® 700 emits at 723 nm, excited with red laser (633 nm). Recommended optical setup: 685 LP mirror, 710/20 filter.
  • Excitation/Emission: 633–647 nm / 723 nm

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 subunits (γ, δ, ε, ζ) are structurally related members of the immunoglobulin superfamily encoded on human chromosome 11.

CD3 plays roles in TCR-induced growth arrest, survival, and proliferation. It is expressed on:

  • 68–82% of normal peripheral blood lymphocytes
  • 65–85% of thymocytes
  • Purkinje cells in the cerebellum

It is not expressed on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
