Product Specific Information
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular biochemical pathways leading to activation and proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse thymocyte and splenocyte suspensions. Recommended ≤0.25 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize concentration for best performance.

Alexa Fluor® 700 emits at 723 nm and is excited by the red laser (633 nm). Most instruments require a 685 LP mirror and 710/20 filter.

Target Information
The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. T cell activation involves the cytoplasmic tails of CD3 gamma, delta, epsilon, and zeta subunits, which associate with signaling molecules via tyrosine-based motifs. CD3 is found on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Reduced T lymphocyte percentages may occur in autoimmune diseases. The genes encoding CD3 epsilon, gamma, and delta are located on chromosome 11. Defects in CD3 are associated with CD3 immunodeficiency.
