Product Specific Information
The OKT3 monoclonal antibody binds to an epitope on the epsilon-subunit of the human CD3 complex. OKT3 has potent immunosuppressive properties and has been effective in treating renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are essential for TCR complex assembly, trafficking, and surface expression. CD3 is expressed by thymocytes during development and by all mature T cells.
Crosslinking of TCR initiates intracellular signaling, leading to activation and proliferation.
This antibody is validated for flow cytometric analysis and has been pre-titrated at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody required to stain cells in a 100 µL volume (typically 10⁵–10⁸ cells/test).
Alexa Fluor® 700 emits at 723 nm and is excited by the red laser (633 nm). Recommended optics: 685 LP mirror and 710/20 filter.

Target Information
CD3 subunit complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits—members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs.
CD3 is present on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Reduced T lymphocyte percentages may occur in autoimmune diseases. Defects in CD3 genes are linked to immunodeficiency.
