For IHC(P), use heat-induced antigen retrieval in citrate buffer (pH 6.0). After incubation with the primary antibody, slides should be incubated with biotinylated secondary antibody, followed by alkaline phosphatase–streptavidin and chromogen.
Target Information
C5AR2 (GPR77) is an orphan member of the anaphylatoxin receptor family. It is a seven-transmembrane G‑protein‑coupled receptor with an N‑terminal N‑glycosylation site, conserved cysteines, and multiple potential C‑terminal phosphorylation sites. It binds C5a and C5a‑des‑Arg, functioning as a regulated scavenger receptor that negatively modulates C5aR‑mediated inflammation. Northern blot analysis shows co‑expression with C5aR in various tissues and cells including neutrophils/macrophages, mast cells, dendritic cells, brain, lung, heart, kidney, liver, ovary, and testis.
