Target Information
KRAB (Kruppel-associated box) 도메인은 약 45개의 아미노산으로 구성된 전사 억제 도메인으로, 다양한 전사 인자에서 발견됩니다. 인간 게놈에는 220개 이상의 KRAB-zinc finger protein (KRAB-ZFP) 유전자가 존재하며, 이들은 KAP-1 (KRAB-associated protein 1)과 상호작용하여 전사 억제를 매개합니다.
KAP-1은 RING finger, B boxes, PHD finger를 포함하는 835 아미노산 길이의 폴리펩타이드로, KRAB 도메인 전사 인자와 복합체를 형성하여 억제 효율을 높입니다. 또한 HP1 및 KRAZ1과 직접 상호작용하며, HP1α가 포함된 중심 이질염색질 부위로 KRAZ1을 표적화하여 전사 억제를 조절합니다.
KRAB 결합은 가능하지만 HP1 결합이 불가능한 KAP-1 변이체는 KRAZ1의 무작위 분포와 강한 전사 활성화를 유도하는 것으로 알려져 있습니다.
