Target Information
The KRAB (Kruppel-associated box) domain (~45 amino acids) is a transcriptional repression domain found in many transcription factors. Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes are identified in the human genome. These proteins repress transcription via interactions with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing RING finger, B boxes, and a PHD finger.
KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency and interacts directly with HP1 (heterochromatin protein 1) and KRAZ1. It targets KRAZ1 to centromeric heterochromatin containing HP1α, regulating transcriptional repression. Mutations disrupting HP1 binding lead to random KRAZ1 distribution and strong transcriptional activation.
