Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription positively or negatively through coactivator or corepressor proteins.
Two receptor forms exist — alpha and beta — encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four estrogen receptor-alpha isoforms (Isoform 1: 66 kDa, Isoform 2: 53 kDa, Isoform 3: 47 kDa, Isoform 4: 35 kDa). Estrogen receptors are broadly expressed in various tissues, essential for sexual development and reproductive function, and also play roles in bone and other tissues. They are implicated in diseases including breast cancer, endometrial cancer, and osteoporosis.
