Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through coactivator or corepressor proteins.
Two forms of estrogen receptors exist—alpha (ESR1) and beta (ESR2)—encoded by separate genes. Alternative RNA splicing produces multiple isoforms of estrogen receptor alpha (66 kDa, 53 kDa, 47 kDa, 35 kDa).
Estrogen receptors are expressed in various tissues and are essential for sexual development, reproductive function, and bone physiology. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
