Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through coactivator or corepressor recruitment.
Two forms exist: ER alpha (ESR1) and ER beta (ESR2), encoded by separate genes. Alternative RNA splicing produces at least four ER-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are broadly expressed and are critical for sexual development, reproductive function, and bone physiology. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
