Target Information
The p16INK4a gene produces multiple transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which are structurally related isoforms functioning as CDK4 kinase inhibitors. Another transcript includes an alternate first exon located 20 Kb upstream, encoding the ARF protein that stabilizes tumor suppressor p53 by interacting with and sequestering MDM2. Despite structural differences, both CDK inhibitor isoforms and the ARF product contribute to G1 phase cell cycle control. This gene is frequently mutated or deleted in various tumors and serves as an important tumor suppressor gene.
