Target Information

Estrogen Receptor beta (ERβ, NR3A2) protein is approximately 55 kDa, with isoforms ranging from 36–59 kDa. Human ERβ was initially cloned from testis. Estrogen receptors belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. When bound to estrogen, ERβ acts as a ligand-activated transcription factor, inducing dimerization and binding to estrogen response elements (ERE) in DNA, thereby regulating gene transcription through coactivator or corepressor recruitment.

Two forms of estrogen receptor exist—alpha (ESR1) and beta (ESR2). ERβ binds estrogens with similar affinity to ERα and can inhibit ERα activity in reproductive tissues. ERβ is widely expressed in many tissues and may function as a tumor suppressor, with roles in prostate cancer, medullary thyroid carcinoma, cardiovascular disease risk, and other conditions.
