Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) present on many stem cell populations. It serves as a stem cell marker with reversible expression on human hematopoietic stem cells. CD34 may function as a surface receptor involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells.
CD34 expression represents a specific state of hematopoietic development with altered adhesion properties and differentiation potential in vitro and in vivo. It may act as an adhesion molecule mediating stem cell attachment to the bone marrow extracellular matrix or stromal cells, and as a scaffold for lineage-specific glycans binding to stromal lectins. CD34 is also implicated in presenting carbohydrate ligands to selectins. The intracellular domain of CD34 can be phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
