Product Specific Information

The monoclonal antibody mhalx21 reacts with mouse interleukin-21 (IL-21), a 17 kDa immunomodulatory cytokine produced mainly by NKT, Th17, and TFH cells.
IL-21 in TFH cells promotes autocrine signaling through IL-21R and STAT3, activating Bcl6 and supporting TFH effector functions. It is essential for T cell-dependent B cell differentiation, IgG production, and induction of apoptosis in naive B cells.

In Th17 cells, IL-21 upregulates IL-23R through STAT3, IRF4, and RORγt, facilitating maturation and maintenance by IL-23, while downregulating Foxp3. Elevated IL-21 is observed in chemically induced colitis models, whereas IL-21-deficient mice show protection from colitis.

Preliminary data indicate that clone mhalx21 recognizes a different epitope than anti-mouse IL-21 clone FFA21 (Product #51-7211).

Applications Reported:
Optimized for intracellular staining and flow cytometric analysis using eBioscience IC Fixation and Permeabilization Kit (Product #88-8823-88).

Applications Tested:
Validated on Th17-polarized splenocytes restimulated with PMA, Ionomycin, and Brefeldin A for 5 hours. Recommended use ≤ 0.25 µg/test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information

IL-21 is a 17 kDa cytokine produced by NKT, Th17, and TFH cells. It mediates autocrine signaling via IL-21R and STAT3, activating Bcl6 and regulating TFH development and function.
IL-21 is involved in B cell differentiation, IgG production, and apoptosis in naive B cells. In Th17 cells, IL-21 drives IL-23R expression and inflammatory maintenance by IL-23, while suppressing Foxp3.
IL-21-deficient mice resist chemically induced colitis, whereas elevated IL-21 levels are observed in colitis models.
