Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta.
The CD8β chain associates with CD8α to form the CD8αβ heterodimer expressed on thymocytes and peripheral cytotoxic T cells. The CD8 receptor complex binds to MHC class I molecules and plays a key role in T cell development and activation.
SIDI8BEE can detect CD8β in both CD8αβ heterodimers and CD8ββ homodimers. Cross-reactivity has been confirmed for human and rhesus macaque CD8β; other primates have not been tested.

Applications Reported: Flow cytometry, immunoprecipitation, and Western blotting.
Applications Tested: Flow cytometric analysis of human peripheral blood lymphocytes at ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration is recommended.

Target Information

The CD8B antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes, mediating cell-cell interactions within the immune system. Acting as a coreceptor with the T-cell receptor, CD8 recognizes antigens presented by MHC class I molecules on antigen-presenting cells.
The functional coreceptor can exist as a homodimer (αα) or heterodimer (αβ). Both chains share homology with immunoglobulin variable light chains. The CD8B gene encodes the β chain isoforms, with multiple alternatively spliced variants and a pseudogene located on chromosome 2.
