Target Information

cGAS (Cyclic GMP-AMP synthase) is a nucleotidyltransferase that catalyzes the formation of cyclic GMP-AMP (cGAMP) from ATP and GTP, playing a crucial role in innate immunity.
Catalysis involves both the formation of 2’,5’ phosphodiester linkage at the GpA step and 3’,5’ linkage at the ApG step, producing c[G(2’,5’)pA(3’,5’)p].
cGAS binds cytosolic DNA directly, leading to activation and synthesis of cGAMP, which in turn activates TMEM173/STING and triggers type-I interferon production.
It is also involved in DNA sensing during infection, stress response, DNA damage detection, and regulation of cellular senescence.
