Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a receptor expressed on activated T cells and occasionally on activated B cells. CTLA-4 binds B7 family members (CD80 and CD86) with higher affinity than CD28, delivering inhibitory signals that limit T cell activation and proliferation.

This antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for intracellular staining and flow cytometric analysis using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) following BestProtocols® guidelines.

For complete CTLA-4 detection, both surface and intracellular expression should be analyzed.

Fluorochrome Details:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass). Ensure instrument compatibility.

Fixation Guidance:
Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) with minimal signal loss.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor primarily expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28 and inhibits T cell activation. Intracellular CTLA4 is also found in regulatory T cells and contributes to their suppressive function.
