Product Specific Information

The mShad150 monoclonal antibody reacts with mouse CD150, an ~70 kDa transmembrane glycoprotein known as Signaling Lymphocyte Activation Molecule (SLAM). CD150 is expressed by T cells (particularly Th1) and B cells, and its expression is rapidly upregulated upon activation. Immature thymocytes and dendritic cells also express this antigen.

Signaling through SLAM in T cells induces proliferation and enhances interferon-gamma response. SLAM may also mediate adhesion between T cells and antigen-presenting cells. The mShad150 antibody stains the CD150⁺CD244⁻CD48⁻CD41⁻ population of pluripotent hematopoietic stem cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested with mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Titrate for optimal performance.

PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5. It emits at 710 nm and is excited at 488 nm (blue laser). Recommended filters: 685 LP dichroic mirror and 710/40 band pass (695/40 acceptable alternative).

PerCP-eFluor® 710 conjugates are stable when fixed with freshly prepared 2% formaldehyde overnight at 4°C.

Target Information

CD150 (SLAM, Signaling Lymphocyte Activation Molecule) is part of the immunoglobulin superfamily and involved in T cell stimulation. It is expressed on peripheral blood memory T cells, immature thymocytes, and certain B cells, and is rapidly induced upon activation.

Activated B cells express membrane-bound, soluble, and cytoplasmic isoforms of SLAM. Homophilic SLAM-SLAM interactions enhance B cell expansion and differentiation. The extracellular domain serves as a receptor for measles virus and acts as a co-activator on T and B cells. CD150 interacts with SH2D1A and PTPN11, and mutations may be associated with X-linked lymphoproliferative disease (XLP).
