Target Information

This gene encodes a member of the tumor necrosis factor (TNF) cytokine family, which acts as a ligand for osteoprotegerin and plays a key role in osteoclast differentiation and activation. It also functions as a dendritic cell survival factor and regulates T cell-dependent immune responses. Activation of T cells induces expression of this gene, leading to increased osteoclastogenesis and bone loss. The protein activates anti-apoptotic kinase AKT/PKB through a signaling complex with SRC kinase and TRAF6, suggesting a role in apoptosis regulation. Gene disruption in mice results in severe osteopetrosis, lack of osteoclasts, and impaired T/B lymphocyte differentiation. Two alternatively spliced transcript variants have been identified.
