Product Specific Information
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is critical for T-cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number may range from 10⁵–10⁸ cells/test. Optimize by titration for best performance.
Fluorochrome: APC-eFluor 780 emits at 780 nm and is excited by a 633 nm red laser. Ensure instrument compatibility.
Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333) for up to 3 days at 4°C in the dark with minimal effect on fluorescence.
Excitation / Emission / Laser: 633–647 nm / 780 nm / Red Laser
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD4 is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed on T helper cells, but also found on monocytes, macrophages, and dendritic cells. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and various tissues including brain and gut. It contributes to T-cell activation via association with the T-cell receptor complex and protein tyrosine kinase Lck. CD4 may also mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
