Product Specific Information

Description:
The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by most thymocytes, a subset of mature T cells (T-helper cells), and at low levels on monocytes. CD4 serves as a receptor for HIV. The OKT4 antibody recognizes a different epitope than the RPA-T4 monoclonal antibody, and these antibodies do not cross-block each other.

Applications Reported:
This OKT4 (OKT-4) antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited with the Red laser (633 nm). Ensure instrument compatibility. Light-sensitive; protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal effect on brightness and FRET efficiency.

Target Information

The CD4 antigen participates in the recognition of MHC class II molecules and acts as a co-receptor for HIV. CD4 is mainly expressed in T-helper cells but also in monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. CD4 aids in T-cell activation through association with the T-cell receptor complex and Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
