Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed on most thymocytes, a subpopulation of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.06 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited with the red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal signal loss. Performance may vary by clone and should be verified empirically.

Target Information

CD4 antigen participates in recognition of MHC class II molecules and acts as a co-receptor for HIV. It is mainly expressed on T helper cells, monocytes, macrophages, and dendritic cells. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues. It contributes to T-cell activation via association with the T-cell receptor complex and Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
