CDK4/cyclin D3 serine/threonine kinase는 세포주기 G1 단계에서 세포 증식 조절에 관여하는 이합체 단백질 복합체입니다.
CDK4는 cyclin-dependent protein kinase 계열에 속하며 D형 cyclin과 복합체를 형성하고, p16(cyclin-dependent kinase inhibitor-2)에 의해 억제됩니다.
CDK4는 Rb 단백질의 C-말단 영역을 인산화하여 E2F 복합체의 전사 억제를 활성화할 수 있습니다.
CDC37 및 HSP90은 D형 cyclin에 결합되지 않은 CDK4와 선호적으로 결합할 수 있습니다.
SMAD3는 CDK4 및 CDK2의 주요 생리학적 기질이며, CDK6과 CDK4는 Rb 단백질의 서로 다른 잔기를 선택적으로 인산화합니다 (CDK6: Thr821, CDK4: Thr826).
