Target Information
Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene. It is a key component of the extracellular matrix in cartilage and provides resistance to compression. The human form (~250 kDa, 2316 amino acids) has multiple isoforms due to alternative splicing.
Aggrecan has a bottlebrush structure with chondroitin sulfate and keratan sulfate chains attached to a protein core. It contains three interglobular domains (IGDs: G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis. Fragments of aggrecan are released into synovial fluid during osteoarthritis progression due to cleavage by aggrecanase and MMP activity. Antibodies BC-3 and BC-4 detect neo-epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
