Target Information
STAT3는 세포 내 신호전달 및 전사 활성화에 관여하는 STAT 단백질군에 속합니다. 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동한 후 전사인자로 작용합니다. CREB-binding protein 등의 공동활성화 인자가 STAT3의 전사 활성에 필요하며, Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화됩니다. Tyr705 위치의 인산화는 JAK1 및 JAK2에 의해 매개되며, 이는 STAT3 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다. 인간 STAT3 유전자는 17번 염색체 q 팔에 위치하며, 세포 성장 및 세포사멸 조절 등 다양한 생리적 과정에 중요한 역할을 합니다. Rac1은 STAT3의 활성 조절에 관여하며, PIAS3는 STAT3의 특이적 억제 단백질로 알려져 있습니다. STAT3는 세 가지 상이한 스플라이싱 변이체를 가지며, knockout 마우스에서는 배아 단계에서 치명적인 결손을 보입니다.
