Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses. PKR plays a key role in antiviral defense through activation by double-stranded RNA produced during viral infection. It is involved in IFN-induced innate antiviral responses, virus-induced apoptosis, and regulation of cell growth and differentiation.

PKR induces apoptosis by up-regulating Fas expression and mediates FADD/Caspase-8 death signaling. Upon viral infection, PKR acts as a dual-function protein, activating both cell survival and death pathways via kinase-independent and dependent mechanisms. It regulates multiple signaling pathways including NF-κB, p53, p38, and PDGF. PKR has been implicated in tumor suppression, malignancy, and several neurodegenerative diseases such as Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis.
