This antibody requires protein digestion pre-treatment of paraffin sections using trypsin or pronase.
A suggested positive control for immunohistochemical applications is liver tissue.
For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.

Mouse anti-Rat CD163, clone ED2, recognizes the rat ED2 cell surface glycoprotein (Dijkstra et al., 1985).

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich (SRCR) superfamily.
It functions as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and contributing to iron recycling through endocytosis of hemoglobin/haptoglobin complexes and heme breakdown.

CD163 is expressed exclusively on monocytes and macrophages, particularly in the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages except those in mantle zones, germinal centers, interdigitating reticulum cells, and Langerhans cells.

Expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding of CD163 into plasma or cell supernatant.
CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F phenotype) than for dimeric haptoglobin (HP1S phenotype).
Binding triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
