Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase family, which includes EGFR, ERBB2, and ERBB3.
ERBB4 is activated by neuregulins and other ligands, inducing cellular responses such as mitogenesis and differentiation.

ErbB4 functions as a receptor for heregulin and mediates HGL-stimulated tyrosine phosphorylation. It plays critical roles in signal transduction regulating metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.

Expression of ErbB4 is high in several breast carcinoma cell lines (e.g., T47-D, MDA-MB-453, BT-474, H3396) and in normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum.
Low or absent expression is observed in some breast tumor cell lines (e.g., MDA-MB-231, MDA-MB-157, MDA-MB-468, SKBR-3). Elevated ErbB4 levels in certain human tumor cell lines suggest a potential role in malignancy.
