Target Information
B7-H4 (also known as B7x, VTCN1) is a costimulatory protein that functions as a negative regulator of T-cell mediated immunity. Although its receptor is unknown, B7-H4 is believed to deliver inhibitory signals to T-cells, preventing proliferation, cell cycle progression, and interleukin-2 production. B7-H4 deficient mice show minimal immune alteration, indicating its role in fine-tuning immune responses.
B7-H4 is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Aberrant expression has been associated with lung, breast, and ovarian cancers. Overexpression may help tumor cells evade immune surveillance by suppressing T-cell activity. VTCN1 is also considered a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
