Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha)는 수용체형 막관통 당단백질로, 과립구, 수지상세포(DCs), 대식세포, 비만세포 및 조혈줄기세포 등 골수계 세포에서 발현됩니다.
SIRP alpha는 활성화된 여러 티로신 키나아제(EGFR, PDGFR, src, insulin receptor 등)의 기질로 작용하며, 수용체 티로신 키나아제 신호 경로의 음성 조절에 관여합니다.
CD47과의 결합은 SIRP alpha의 세포질 내 ITIM 영역의 인산화를 유도하고, SHP-1 및 SHP-2 티로신 인산화효소의 활성화를 매개합니다.
이 상호작용은 대식세포의 식세포 작용 억제, T 세포 및 DC의 양방향 활성화 등 다양한 면역 조절 과정에 관여합니다.
암세포는 CD47을 과발현하여 SIRP alpha를 활성화시키고, 대식세포에 의한 암세포 제거를 억제합니다.
