Product Specific Information
The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells, playing a crucial role in T cell development and activation. Binding of RM4-5 is blocked by GK1.5.
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated for flow cytometric analysis of mouse thymocytes and splenocytes at ≤0.5 µg/test (100 µL final volume).
Usage Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- Shipments include CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining performance.
- Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.

Target Information
CD4 is a co-receptor for MHC class II recognition and HIV binding, primarily expressed on T helper lymphocytes, but also found in monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and specific brain and gut regions. CD4 facilitates early T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple splice variants have been identified (RefSeq, July 2017).
